      Abstract
      Vanda tricolor orchid becomes an endemic orchid of Mount Merapi, which grows around the slopes of Mount Merapi. This orchid has a longer growth response compared to other orchids in general. The combination of medium and ZPT in in vitro culture propagation will affect the growth of the orchid. To determine the type of media and concentration of ZPT cytokinin (TDZ) suitable for growth of PLB Orchid Vanda tricolor, the research was arranged in a Factorial Randomized Design (RAL) 3x3 with 2 factors, the first factor was media (MS, VW, and NDM) factors second is Thidizuron concentration (0 mg/L, 0.5 mg/L and 1 mg/L). Research conducted at the Laboratory of Culture In vitro Faculty of Agriculture, University of Muhammadiyah Yogyakarta from January to April 2018. The results showed that PLB gave a growth response to the treatment given, although the analysis showed no interaction or no significant difference in diameter increasing parameters, time of buds, buds number and time of buds appeared. The use of PLB (protocorm like bodies) as eksplan resulted in high percentage of life explant (100%) and no occurrence of browning or contamination on explant. The NDM medium tends to give a higher rate of diameter PLB (1.12 mm), higher percentage of PLB leaves appear (60%), and faster root time (0.90 weeks). Concentration of 0.5 TDZ mg /L tends to give larger diameter increment (1.02 mm), faster shoots appear (1.46 weeks), total shoots more (1.80 shoots), and rooted explant percentage higher (70%).
