      Abstract
      Red betel plant is Indonesia’s original plant which has many benefits on health care. Empirically, red betel has been widely used by people as a traditional medicine. One of them used as a cure wounds and antiseptic. The purpose of this study was to develop an antiseptic gel formulation using ethanolic extract of red betel leaf as the active ingredient. The extraction was conducted by maceration method using ethanol 70% as the solvent. Carbomer was used for the gelling agent and the extract concentration that was tested are 0%, 2,5%, 5%, 10% and 15%. The effectiveness test of antiseptic gel was using replica method by testing after and before applying the gel on the hand. The antiseptic gel was droped on the clean hand and than swabbed using cotton bud than inoculate on the agar media. Further, the agar than incubated in the incubator for 24 hours at 370C. The evaluation was conducted by calculating the number of colony of the bacteria. Gel Antiseptic C was used as the positive control. Then statistical tests between positive control and F1, F2, F3, F4, F5 were using SPSS 16.0. The results of the replica test before and after applays the antiseptic red betel extract leaf with levels of 0%, 2.5%, 5%, 10% and 15%, obtained a decrease in the total number of germs, i.e 1,38% ± 0.29, 2,89% ± 1.04, 27,19% ± 3.59, 68,10% ± 7.44 and 85,62% ± 6.59, respectively. The decline of the total number of germs resulting from the positive control of gel "C" are 99.14% ± 0.46. The F1, F2, F3, F4, F5 results obtained significant difference (p<0,05) compare to positive control. It can be concluded that the red betel leaf antiseptic gel effective as antiseptic hand, although the antiseptic effectiveness is still lacking when compared with antiseptic preparations commercially available.
