PENGARUH PERANCAH HIDROGEL CaCO3 DENGAN INKORPORASI PLATELET RICH PLASMA TERHADAP WAKTU BERHENTINYA PERDARAHAN PADA LUKA TIKUS PUTIH (Rattus norvegicus)
Abstract
Background : Bleeding is a normal body response after tooth extraction. Bleeding can be a dangerous condition if it occurs excessively. Excessive bleeding after dental treatment can cause patients to be shocked, syncope, and even death if not treated properly. Effective hemostatic agents are needed to prevent excessive bleeding. Various types of hemostatic agents have been developed by researchers and used by practitioners in their clinics. The purpose of this study was to determine the effect of CaCO3 hydrogel scaffold with incorporation of Platelet Rich Plasma (PRP) to the cessation time of bleeding in the wound of white rats (Rattus norvegicus).
Method : The research method is an experimental laboratory with post test design. This study had 4 treatment groups, namely the first group used CaCO3 hydrogel scaffold with incorporation of Platelet Rich Plasma, the second group used only CaCO3 hydrogel scaffold, the third group used hemostatic Spongostan agents as a positive control, and the last group without treatment or negative control. Each treatment group consisted of 6 research rat samples, then the total sample used was 24 rats. Rat tail was cut 2 cm from the tip and then treated according to the sample groups. Measurements the cessation time of bleeding starts from blood first dripping until there is no more blood spots that come out and imprint on filter paper. Measurement the cessation time of bleeding using a stopwatch device.
Result : The results showed that the average cessation time of bleeding in CaCO3 hydrogel scaffold group with incorporation of PRP was 177, 33 seconds, the CaCO3 scaffold group was 314.67 seconds, Spongostan group was 276 seconds, and the negative control group was 387.83 seconds. The One Way ANOVA statistical test results showed a significant difference (p <0.05) between the CaCO3 hydrogel scaffold group with the incorporation of PRP with a group without a treatment or negative control. Conclusion : The CaCO3 hydrogel scaffold with incorporation of Platelet Rich Plasma has the shortest cessation time of bleeding and has a significant effect in shortening the time of cessation of bleeding compared to injury without the treatment of any hemostatic agent.
