dc.description | Polyvinyl alcohol (PVA) dan kitosan merupakan perpaduan polimer yang biasa digunakan sebagai bahan pembalut luka (wound dressing) yang dapat dikembangkan dalam bentuk membran nanofiber karena memilki sifat biocampatible, biodegradable dan tidak beracun (non toxic).Electrospinning merupakan metode yang paling mudah dan banyak digunakan dalam pembuatan membran nanofiber. Membran PVA/kitosan yang digunakan sebagai pembalut luka harus memiliki sifat mekanis native skin dengan nilai kuat tarik antara 5-30 MPa dan modulus elastisitas antara (4,6-20 MPa). Tujuan penelitian ini adalah mengetahui pengaruh penambahan kitosan nanoemulsi (NeCs) terhadap morfologi dan sifat mekanis membran nanofiber PVA/NeCs. Selain itu, mengetahui efektivitas membran PVA dan PVA/NeCs terhadap penyembuhan luka sayat mencit.
Proses electrospinning dilakukan pada tegangan 11 kV, diameter jarum syringe 0,6 mm dan jarak ujung jarum dengan kolektor (TCD) 12 cm. Larutan PVA 10% (w/w) digunakan sebagai matriks dengan penambahan 0%, 5%, 10% dan 15% (w/w) NeCs. Sifat fisis (konduktivitas dan viskositas) larutan polimer PVA dan PVA/NeCs diuji menggunakan konduktometer dan viskometer. Morfologi membran dikarakterisasi menggunakan scanning electron microscope (SEM), dan sifat tariknya menggunakan Universal Testing Machine (UTM). Uji in vivo luka dilakukan selama 17 hari menggunakan membran PVA/NeCs yang dibandingkan dengan pembalut komersial (Sofra-Tulle) dan diujikan pada luka mencit yang dibagi kedalam 3 kelompok. Kelompok I (PVA murni), kelompok II (PVA/NeCs 15%/perlakuan) dan kelompok III (Sofra-Tulle).
Hasil penelitian menunjukkan bahwa penambahan NeCs pada larutan PVA/NeCS menurunkan nilai viskositas, tetapi meningkatkan konduktivitas larutan. Sifat tarik membran PVA/NeCs yang dibuat menggunakan PVA low molecular weight mendekati sifat mekanis native skin.. Nilai kuat tarik tertinggi didapat pada membran PVA/NeCs 5% (13,68 MPa) dan terendah terdapat pada membran membran PVA/NeCs 15% (8,27 MPa). Modulus elastisitas meningkat dari 36,69 MPa menjadi 59,70 MPa. Sedangkan untuk uji efektivitas penyembuhan luka menunjukkan bahwa membran PVA/CsNe 15% mampu mendekati kemampuan pembalut luka komersial. | en_US |
dc.description.abstract | Polyvinyl alcohol (PVA) and chitosan are a combination of polymers commonly used as a wound dressing because they have biocompatible, biodegradable and non-toxic properties that can be developed to be the nanofiber membranes. Electrospinning is the easiest method and widely used in making nanofiber membranes. PVA/Chitosan membrane used as wound dressing must have native skin mechanical properties, i.e. high tensile strength (5-30 MPa) and medium modulus of elasticity (4.6-20 MPa). The purpose of this study was to determine the effect of adding chitosan nanoemulsion (NeCs) to on the morphology and mechanical properties of PVA/NeCs nanofiber membranes. Moreover, knowing the useful of PVA and PVA/NeCs membranes on wound healing in mice wounds.
The electrospinning process was carried out at a voltage of 11 kV, the diameter of the syringe needle was 0.6 mm and the distance of the needle tip with the collector (TCD) was 12 cm. 10% PVA solution (% by weight) is used as a matrix mixed with NeCs using concentrations of 0%, 5%, 10% and 15% (weight%). The physical properties of the solution were tested using a conductometer and viscometer. Membrane morphology was observed using Scanning Electron Microscope (SEM), then examined its tensile properties using Universal Testing Machine (UTM). In vivo wound tests were carried out for 17 days using PVA / NeCs membranes compared to commercial wound dressing (Sofra-Tulle) and tested on mice wounds divided into 3 groups. Group I (pure PVA), group II (PVA / NeCs 15% / treatment) and group III (Sofra-Tulle).
The results showed that the addition of NeCs decreases the viscosity, but it increases the conductivity of the PVA/NeCS solution. Tensile properties of PVA / NeCs membranes made using PVA low molecular weight approached native skin mechanical properties. The highest tensile strength was obtained from PVA/NeCs 5% membrane (13.68 MPa) and the lowest was found in PVA/NeCs 15% membrane (8.27 MPa). Modulus of elasticity increased from 36.69 MPa to 59.70 Mpa, whereas the effectiveness of wound healing test showed that 15% PVA/CsNe membrane was able to approach the commercial wound dressing capability | en_US |
