      REGENERASI ANGGREK Vanda tricolor PASCA ERUPSI MERAPI MELALUI KULTUR IN VITRO

      Abstract
      The eruption of the Merapi volcano and the exploitation of the orchid from its habitat had decreased the population of Vanda tricolor. The conservation of the orchid could be done through in vitro culture. The aim of the research was to determine the best of media and concentration of Benzyl Amino Purine, Naphtalene Acetic Acid and Thidiazuron on the callus growth of Vanda tricolor. The research was consisted of two experiments. First experiment was arranged in a completely randomized design factorial with nine treatments. The treatments were combination between media type (1/2 MS and NDM) and the concentration of BAP (0, 1, 2, 3, 4 mg/l) and NAA (0; 0,1; 0,5 mg/l). The second experiment was arranged in a completely randomized design, single factor, with six treatments (½ MS media + 0 mg/l Thidiazuron, ½ MS media + 0.5 mg/l Thidiazuron ½ MS media + 1 mg/l Thidiazuron, NDM media + 0 mg/l Thidiazuron, NDM media + 0,5 mg/l Thidiazuron, and NDM media + 1 mg/l Thidiazuron. Each treatment was repeated 15 times, each replication consisted of one sample. The results showed that NDM media supplemented with 1 mg / l BAP + 0.1 mg / l NAA and 2 mg / l BAP + 0.1 mg / l NAA induced the callus of Vanda tricolor from ex vitro leaf explants. NDM Media supplemented with 0,5 mg/l Thidiazuron was the best treatment to induce the callus of Vanda tricolor from in vitro leaf explants.
